Malachite green (MG) is a triphenylmethane dye, which is used extensively in aquaculture for the treatment or prevention of external fungal and parasitic infections in fish as well as their eggs. MG is rapidly absorbed by fish and fish eggs during waterborne exposure. The reduced derivative leucomalachite green (LMG) is recognized as a major metabolite of MG in fish tissues. Fish treated with malachite green will excrete MG rapidly, but will store LMG in muscle tissue for months. The use of MG in aquatic food animals is highly restricted or banned in several countries because of toxicological considerations. It has been reported to cause carcinogenesis, mutagenesis, chromosomal fractures, tetragenecity and respiratory toxicity in animals.
According to the European Commission, methods that can be used for the determination of MG residues in fish muscles must meet the required performance limit of 2ug/Kg for the summation of MG and LMG. Several analytical methods have been described for Malachite Greens (MGs) detection, such as HPLC and LC-MS. These techniques require expensive apparatus and reagents, in addition to being time-consuming.  A more sensitive, rapid and inexpensive method for detection of MGs is needed.
An indirect competitive ELISA kit was offered to efficiently detect LMG , reduced MG or other drugs of triphenylmethane dye such as crystal violet (CV) or leucocrystal violet (LCV) residues in samples by TABP. The detection limit for MGs is 0.2 ppb, and there is no cross reaction to other antibiotics and sulfa drugs. The MGs ELISA kit is an enzyme.
